Protein phosphorylation is a highly important mechanism in cell signalling pathways for regulating protein function. In particular phosphorylation is the reversible addition of a phosphate group to a protein or other organic molecule resulting in a conformational change in the structure and causing activation or deactivation of protein’s function. Analysis of protein phosporylation is problematic as the concentration of any given phosphoprotein and the ionization efficiency of phosphopeptides for MS are relatively low. GL Sciences’ Titanium Dioxide (TiO2 or Titania) products offer a new efficiecy level in the field of purification and enrichment of phosphopeptides replacing IMAC as the primary sample preparation method prior to LC/MS analysis.
GL Sciences’ produces Titanium Dioxide as highly spherical beads with the optimum ratio of crystal forms of TiO2. This unique formulation of titanium dioxide beads lead to a superior performace of GL Sciences’ Phos-TiO and MonoTip products.
Under acidic conditions phosphate groups are preferentially adsorbed to the surface of TiO beads are eluted under basic conditions.
Titansphere TiO bulk sorbent media is available in particle sizes 5µm und 10µm in quantities of 500 mg. The Titanium Dioxide crystals are spherical with a pore size of 100Å. They are used in a pH range of 2-12.

The Titansphere Phos-Tio Kit contains tip-columns with titansphere media in the tip. The convenient 200µl tip provides 3 mg Tip material also offered as bulk and the 10 µl size tip 1 mg TiO. The kits also include waste and collection tubes as well as all required buffers.

Styrene divinylbenzene SDB and graphite carbon GC centrifuge-operated micropipette GL-Tips allow a higher retention of a wider range of phosphopeptides. GL Sciences GL-Tip SDB is more hydrophobic than C18 and GL-Tip GC retains more hydrophilic phosphopeptides. They are the ideal TiO-2 desalting products prior to LC/MS.
